Improved version of the Kean partition assay for cerebroside sulfate.

چکیده

In the Kean method for the colorimetric determination of sulfatide (1968. J. Lipid Res. 9: 319-327), the lipid is partitioned together with a blue cationic compound between two phases formed from chloroform, methanol, and water. The blue cation enters the chloroform-rich phase only as an ion pair with the lipid. This method has been improved by the use of a new mixture of solvents in which the desired layer floats above the excess dye. The lower volatility of the new solvent system improves the reproducibility of the technique.

Brain Lipids: I. Quantification and Fatty Acid Composition of Cerebroside Sulfate in Human Cerebral Gray and White Matter.

Cerebroside sulfate was isolated by column chromatography from infant whole brains and adult human cerebral grey and white matter. Fatty acid analyses, performed by gas-liquid chromatography (GLC), of normal and ahydroxy fatty acids obtained after hydrolysis revealed that cerebroside sulfate contains long-chain fatty acids ranging from 14-26 carbons both in the normal and hydroxy series. In mat...

The fatty acid composition of brain sphingolipids: sphingomyelin, ceramide, cerebroside, and cerebroside sulfate.

Sphingomyelin, ceramide, cerebroside, and cerebroside sulfate from beef brain were isolated by column chromatography in quantitative yield and in pure form. The fatty acid composition of each sphingolipid was determined by gas-liquid chromatography (GLC). Sphingomyelin contained only nonhydroxy fatty acids, while cerebroside, cerebroside sulfate, and ceramide contained both nonhydroxy and hydro...

Short chain ceramides as substrates for glucocerebroside synthetase. Differences between liver and brain enzymes.

In order to increase the sensitivity of the assay for ceramide: UDPGlc glucosyltransferase, the enzyme that makes glucocerebroside, we synthesized a variety of ceramide homologues that might be better substrates than the naturally occurring ceramides. N-Octanoyl sphingosine proved to be the best lipid tested in liver and brain. It could be added to the tissue homogenate in the dry form, as a th...

A novel mass spectrometric assay for the cerebroside sulfate activator protein (saposin B) and arylsulfatase A.

A mass spectrometric method is described for monitoring cerebrosides in the presence of excess concentrations of alkali metal salts. This method has been adapted for use in the assay of arylsulfatase A (ASA) and the cerebroside sulfate activator protein (CSAct or saposin B). Detection of the neutral glycosphingolipid cerebroside product was achieved via enhancement of ionization efficiency in t...

Cerebroside sulfate (sulfatide A) in some organs of the rat and in a mast cell tumor.

A noteworthy characteristic of cerebroside sulfate (2) (sulfatide A (3)) is its continual accumulation in the brain of rats (4), human beings (5), and mice (6). Recent experiments (7) with radioactive galactose and sulfate showed that cerebroside sulfate of rat brain, unlike other galactolipids, turns over slowly, if at all. Sulfolipids have been described in organs other than brain, including ...
